I've been using an app on my phone while looking for hookups. I met a guy and through the chats I am 99% sure I know this guy in real life. We've had a really good conversation, but there's no way we could ever hookup. He's a supervisor at my company, but not mine. At work we interact a couple times a week. How do I let him know I'm not interested?

Always be honest. Simply tell him that you may know each other and probably shouldn't get together. Honesty is a personal virtue that is important to keep, when it is easy or convenient to stray from it you cannot claim to be or expect to be reputed as honest.
